Measurement of Staphylococcus aureus Phagocytosis by Human Leukocytes -Comparison of Flow Cytometry with Immune Microscopy-

|
Phagocytosis of bacteria by neurophilic polymorphonuclear leukocytes and macrophages represents a cornerstone of host defence against bacterial infection. To determine the level of phagocytosis for neutrophils, many studies are available such as
latex
beads, icdrobiologic methods and radio isotopes. The present study was to compare the results of S. aureus phagocytosis measured by flow cytometry with those by immunofluorescent microscopy. The neurtophils from 10 healthy volunteers incubated
with
fluorescence tagged. S. aureus in human serum and washed in buffer to remove the extracellular bacteria. These cell were analyzed by flow cytometry and immune microscopy. The rate of phagocytosis of flow cytometry was 69.4% and that by
immunofluorescent
microscopy was 62.9%. The comparison revealed the correlation in high agreement (r=0.802). In conclusion flow cytometry facilitated a reliable and rapid in vitro evaluation of S. aureus phagocytosis of polymorphonuclear leukocytes.
